摘要
应用戊二醛法偶联HM-3与BSA,合成完全抗原,免疫新白兰白兔,制备HM-3多克隆抗体。同位素示踪法测定多抗效价、特异性和亲和力。结果表明:抗体的效价为1∶2000;亲和力常数经Scatchard分析计算为2.84×109L/mol,达到高敏分析要求;交叉实验结果表明抗体有较高的识别特异性。HM-3多抗的制备为放射免疫分析方法的建立奠定了基础。
In order to prepare the polyclonal antibody of HM- 3, glutaral coupling was used to couple HM- 3 with the carried protein BSA to obtain synthetic antigen. The anti - HM- 3 serum was produced by immunizing New Zealand rabbits. Polyclonal antibody's valence, specificity and affinity were detected by Isotope tracer meth- ock The results showed that the molecular conjugate ratio between BSA and HM- 3 is 1 : 30. The antibody possessed titer of 1 : 2000 and affinity constant(ka) of 2. 84× 10^9L/mol according to Scatchard analysis, which was quite adequate; cross reaction test showed high specificity. This has laid the basis for RIA of HM- 3.
