本文研究了金蝉口服液体内外抗流感病毒的活性。体外采用细胞病变效应(CPE)观察金蝉口服液对MDCK细胞的毒性浓度;采用血凝法观察金蝉口服液对流感病毒A/FM1/1/47(H1 N1)的抑制活性,并比较预防法、直接灭活法Ⅰ、直接灭活法Ⅱ和治疗法处理病毒时的效果差异。体内采用ICR小鼠滴鼻感染流感病毒造成肺炎模型,以小鼠死亡率和死亡保护率为指标,观察金蝉口服液体内对流感病毒的抑制作用。结果表明,高浓度金蝉口服液对MDCK细胞具有毒性作用,其最大无毒稀释度为1∶128;不同药物处理方式体外对病毒的抑制效果不同,直接灭活法Ⅰ和直接灭活法Ⅱ对该病毒毒株的抑制效果较为明显,其半数有效稀释倍数分别达1∶686和1∶740,而预防法未见明显效果。金蝉口服液对病毒性肺炎引发的小鼠死亡具有保护作用,且以2.0 ml/kg剂量组效果最佳。
This research intended to study the antiviral activity of Jinchan oral solution against influenza virus in vitro and in vivo. Toxicity concentration of Jinchan oral solution to MDCK cells was assessed by observation of cytopathic effect (CPE). Its inhibitory activity against infiuenza A/FM1/1/47/(H1 N1) virus in vitro was determined by hemagglutination assay, and the antiviral activity was compared by using different methods. Viral pneumonia model in ICR mice was used in vivo. The death rate and death protection rate were detected to evaluate the anti-influenza viral effect of Jinchan oral solution. The results showed that Jinchan oral solution had no toxicity to MDCK cells in 1 : 128 dilution. The 50G inhibitory dilution of 1 : 686 and 1 :740 was calculated when the cells were treated in direct deactivation delivery Ⅰ and direct deactivation delivery Ⅱrespectirely, while no inhibitory activity was found when the cells were treated in precaution delivery. Jinchan oral solution displayed the protective effect against influenza virus in infected mice at the dose of 2. 0ml/kg.
