摘要 目的:研究转化生长因子β1(TGF-β1)对人T淋巴细胞的增殖及活化的影响。方法:以MTT法检测TGF-β1对T淋巴细胞增殖的影响;流式细胞术检测TGF-β1对T淋巴细胞活化的影响及CD4+CD25+调节性T淋巴细胞的数量;ELISA法检测TGF-β1对T淋巴细胞分泌干扰素-γ(IFN-γ)、白介素-2(IL-2)的影响。结果:不同浓度的TGF-β1分别作用不同时间后,均可抑制T淋巴细胞的增殖,并随时间的延长、浓度的增加抑制作用增强。不同浓度的TGF-β1作用72h,均能抑制T淋巴细胞活化,活化标志CD69表达率明显降低(P<0.01);均能明显上调CD4+CD25+调节性T淋巴细胞的比例(P<0.01);均能抑制T淋巴细胞分泌IFN-γ、IL-2(P<0.01),且呈剂量依赖性。结论:TGF-β1可以抑制T淋巴细胞的增殖,抑制T淋巴细胞活化,上调CD4+CD25+调节性T细胞的比例,并抑制IFN-γ、IL-2的分泌。 Objective To explore the effects of transforming growth factor β1 (TGF- β1) on the proliferation and activation of T lymphocytes in titro. Methods Effect of TGF- β1 on the proliferation of T lymphocytes was detected by MTI', the effect on the activation of T lymphocytes and the number of CD4+CD25+ regulatory T lymphocytes were measured by flow cytometry (FCM), and the effect on the secretion of interferon- γ (IFN-γ) and interleukin-2 (IL-2) in T lymphocytes was detected by ELISA. Results TGF- β1could inhibit the proliferation of T lymphocytes, and this effect was reinforced with the increase of time and concentration. After being treated with different concentrations of TGF- β1 for 72 h, the activation of T lymphocytes were inhibited, the expression of CD69 was decreased (P 〈 0.01 ), the number of CD4+CD25+ regulatory T lymphocytes was increased (P 〈 0.01 ), and the secretion of IFN-γ and IL-2 were inhibited (P 〈 0.01 ), and all the effects mentioned above were in a concentration dependent manner. Conclusion TGF-β1 can inhibit increase the level of CD4+CD25+ regulatory T lymphocytes, and can inhibit the secretion of of IFN-γ and IL-2.
