654-2在肝缺血再灌注对bcl-2蛋白的影响

Protective Effectiveness of 654-2 on Hepatic Ischemia-Reperfusion in Rats

摘要 目的探讨654-2对肝缺血再灌注损伤(I-R)的保护作用及对bcl-2蛋白表达的影响机制。方法40只Wistar大鼠随机分为I-R组和654-2组,I-R模型由夹闭门静脉左支45min再开放3h制备,比较两组动物血清ALT、GSH和肝细胞bcl-2蛋白水平变化。结果654-2组ALT指标显著低于I-R组(P<0.01),而GSH含量显著高于I-R组654-2组(P<0.01),bcl-2蛋白表达明显高于I-R组(P<0.01)。结论654-2可通过上调肝细胞bcl-2蛋白的表达而对肝缺血再灌注损伤(I-R)有保护作用。 Objective To study the protective effectiveness of 654- 2 on hepatic ischemia- reperfusion injury in rats. Methods Fourty male Wistar rats were randomly assigned into the control group (n =20) and preconditioning group (n = 20)(654 -2 group). The animals were injected 654 -2 in portal vein. After 45 minutes -closure of the animals'left portal veins, blood sample, hepatic tissue regular- dye and bcl - 2 dye were studied 3 hours after the re - open. Results In 654 - 2 group ALT were significantly decreased. Microscopic bcl -2 dye and their GSH of serum were singnifieantly increased. Conclusion It is suggested that 654-2 has protective effect on hepatic ischernia - reperfusion injury.
