川芎嗪抑制内毒素致血管内皮细胞纤溶酶原激活物抑制剂的表达 被引量:14

摘要 纤溶酶原激活物抑制剂1(PAI-1)是一种重要促凝血因子。内毒素脂多糖(LPS)通过刺激血管内皮细胞(EC)分泌PAI-1且而导致血栓形成。采用ELISA的Northern印迹方法观察中药川芎嗪对EC及LPS致EC表达PAI-1的影响。结果表明川芎嗪不仅抑制LPS致EC PAI-1的分泌和mRNA表达,而且抑制 EC PAI-1基础水平mRNA表达。川芎嗪对LPS致 EC PAI-1分泌的抑制呈剂量效应关系。为川芎嗪抗感染性休克和治疗心血管疾病提供了重要的理论和实验依据。 Plasminogen activator inhibitor 1 (PAI-1) is one of important coagulant factors. Endotoxin lipopolysaccharide (LPS) induces thrombosis by stimulating PAI-1 secretion of vascular cells (EC). Using sandwich enzyme-linked immunosorbent assay (ELISA) and Northern blot, the effects of Chinese medicine ligus-trazini on PAI-1 expression in EC and LPS-stimulated EC were studied. The results showed that ligustrazini inhibited both basal and LPS-induced PAI-1 mRNA expression in EC. The effect of ligustrazini on LPS-induced PAI-1 secretion was in a dose-dependent manner. This work provided a theoretic and experimental evidence for ligustrazini against septic shock and cardiovascular disease.
