善胃Ⅰ号方对胃癌前病变大鼠胃粘膜NHE1蛋白及胃内pH值的影响 被引量:2

摘要 目的:通过观察善胃Ⅰ号方对胃癌前病变模型大鼠胃粘膜NHE1蛋白及胃内pH值的影响,从胃内环境角度,探讨该方干预胃癌前病变的作用机制。方法:将80只健康Wistar大鼠随机分为4组,即正常对照组、模型对照组、阳性药物组和善胃Ⅰ号方组,每组20只,造模24周后开始给予药物干预,给药6周后,采用免疫组化法检测大鼠胃粘膜NHE1蛋白的表达水平,高精度酸度计测定大鼠胃液pH值的变化。结果:善胃Ⅰ号方组NHE1蛋白的阳性表达率及胃液pH值较模型对照组及阳性药物组均明显降低(P<0.05)。结论:善胃I号方可能通过抑制胃癌前病变模型大鼠胃粘膜NHE1蛋白的表达,降低胃液pH值,有效维持胃内酸性环境,从而起到干预胃癌前病变发展的作用。 Objective: To study how dose Shanwei I change the expression of NHE1 prote and the PH value in gastric juice, by detecting the effect of Shanwei I on the expression of Na + - H + exchanger-1 ( NHE1 ) and the value of pH in gastric juice in gastric precancerous lesion in rats with blood stasis and beat-toxin. Methods: The 80 rats were divided into four groups: blank control group, model group, positive medicine group and Shanwei I treatment group. After 24 weeks of drug treatment, positive control group and Shanwei I treatment group have to get treatment for 6 weeks. And then immunohistochemistry analysis was used to detect the expression of NHE1 protein in gastric precan- cerous lesions. Results: The expression of NHE1 protein in the gastric mucosa and the value of pH in gastric juice of the rats in Shanwei I treatment group' s expression was significantly lower than other groups. ( P 〈 0. 05 ). Conclusion: Shanwei I treatment group can effectively maintain the intragastric environment acidity, and reduce the NHE1 protein expression in the gastric mucosa.
