Vitamin D is one of the primary regulators of Calcium homeostasis in the body and in criticaly important for the normal mineralization of bone. l, 25(OH) 2D 3 is produced by sequential hydroxylations of vitamin D in the liver and in the kidney. The Classical target organs of 1, 25 (OH) 2D 3 are intestine,bone and kidney. A plethora of tissues to possess receptors to 1, 25 (OH) 2D 3. Vitamin D has many additional functions, such as immunosuppression, stimulates cell differetiation. and inhibits cell proliferation. The VDR genotype hypothesis remains controversial, using subjects of different ethnic background, have or not found a similar correlation hetween the different VDR alleles and bone density. Many 1, 25 (OH) 2D 3 analogs have heen synthesized , some of which were found to be more potent than 1, 25 (OH) 2D 3, at inhibiting tumor growth and less likely to induce hypercalcemia.
