摘要
目的:研究外源性人甲状旁腺素(134)(hPTH(134))对原代培养Wistar新生鼠成骨细胞增殖作用的影响及蛋白激酶A(PKA)、蛋白激酶C(PKC)在此过程中作用。方法:Wistar大鼠成骨细胞原代培养,3HTdR掺入法测细胞增殖。结果:108mol/LhPTH(134)在以24h为一循环的前12h给药循环3次时3HTdR掺入较对照组增加32%,而107mol/LhPTH(134)对3HTdR掺入无影响。hPTH(134)72h持续作用组3HTdR掺入与对照组相比无明显差异。加入PKA抑制剂后阻断hPTH(134)引起3HTdR掺入增加,加入PKC抑制剂后部分阻断108mol/LhPTH(134)引起3HTdR掺入增加。hPTH(134)在1011~108mol/L范围内刺激细胞增殖与浓度成正相关。结论:PTH刺激成骨细胞增殖与给药方式及药物浓度有关,低浓度间歇性给与PTH能刺激成骨细胞增殖。
To investigate the effect of Parathyroid hormone (PTH) on proliferation of osteoblasts and the effect of protein kinase A (PKA) and protein kinase C (PKC) in this process, we isolate the osteoblasts from newborn Wistar rat and count 3H thymidine incorporation radioactivity. The result indicated that 10 9 、10 8 mol/L hPTH(1 34) significantly increase the 3H thymidine incorporation for the first 12h of each 24h incubation cycle for 3 cycles (P<0.01). 10 7 mol/L hPTH (1 34) treated the cells by this manner had no effect compared with the control (P>0.05). While given the osteoblasts with 10 8 mol/L hPTH (1 34) for 72h continuously, the 3H thymidine incorporation had no discrepancy compared with the control. PKA inhibitor blocked the effect of PTH on osteoblasts, while PCK inhibitor partially block this process. From 10 11 to 10 8 mol/L, the effect of PTH on osteoblast′s proliferation is positively relative to its concentration. So we could say, PTH has diverse effect on osteoblasts proliferation depending on the exposure time and its concentration in vitro, and PKA signal transduction systems had the major effect in this process.
