循环高速逆流色谱从银杏叶提取物中制备异鼠李素和山奈酚 被引量:4

Preparation of Kaempferol and Isorhamnetin from Ginkgo Leaves by Recycling High-Speed Counter-Current Chromatography

摘要 为研究分离异鼠李素和山奈酚的方法,制备两者纯品.银杏叶经乙醇提取、氯仿萃取、D101大孔吸附树脂纯化后得到银杏黄酮粗提物,粗提物再经循环高速逆流色谱(循环HSCCC)分离结构类似的异鼠李素和山奈酚,溶剂体系为正己烷∶乙酸乙酯∶甲醇∶水(5∶5∶5∶5).结果表明:粗提取经循环HSCCC循环3次后,异鼠李素和山奈酚达到很好的分离,得到了两者的单体,纯度均在97%以上. Objective: To establish a good procedure for separating kaempferol and isorhamnetin and to gain the pure sample of the two compounds. Methods: Ginkgo (Ginkgo biloba) leaves were extracted by alcohol and then purified by chloroform extraction and D101 macroporous resin to obtain the crude extract of flavone. Then the recycling high-speed counter-current chromatography (recycling HSCCC) was used to separate kaempferol and isorhamnetin from the crude extract with a solvent system composed of n-hexaneethyl acetate-methanol-water (5 : 5 : 5 : 5, v/v/v/v). Results: Kaempferol and isorhamnetin achieved high resolution by recycling HSCCC preparation with three cycles and the purity of the two compounds prepared by recycling HSCCC were over 97 ~. Conclusion: The recycling HSCCC was capable of efficiently separating compounds similar in chemical constitution, which provides a convenient and rapid way to separate kaempferol and isorhamnetin from ginkgo leaves.
