摘要 目的建立大蓟中绿原酸的含量测定方法。方法采用高效液相色谱法测定,色谱柱为Dikma Kromasil C18柱(250mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸(12∶88),检测波长326nm,柱温30℃,流速1.0mL/min,进样量10.0μL。结果绿原酸进样量在0.12~1.2μg范围内与峰面积线性关系良好,r=0.9999,平均回收率为98.13%,RSD为1.23%(n=6),大蓟中绿原酸的平均含量为1.3609mg/g。结论HPLC法简便易行、快速准确,具有良好的重现性和回收率,可作为大蓟中绿原酸的定量分析方法。 Objective To establish a method for the content determination of cblorogenic acid in Cirsium Japonicum. Methods The content of chlorogenic acid was detected by HPLC. The separation was carried out on the Dikma Kromasil C18 column (250 mm × 4. 6 mm, 5 μm). The mobile phase was acetontrile-0. 1% phosphoric acid solution(12: 88). The flow rate was 1.0 mL/min. The detective wavelength was set at 526 nm. The column temperature was 30 ℃ and the sample size was 10. 0 μL. Results The calibration curve showed the good linearity over the range of 0. 12 - 1.2 μg( r = 0. 999 9). The average recovery rate was 98.13% with RSD 1.23% ( n = 6). The sample contained chlorogenic acid 1. 360 9 mg/g. Conclusion This method is simple, rapid and accurate with good repeatability and recovery, which can be used as the quantitative analysis method for Cirsium Japonicum.
