Effect of Visfatin on Secretion and Expression of Inflammatory Factors in SW872 Adipocytes

摘要 目的观察visfatin对SW872成熟脂肪细胞炎性因子TNF-α、单核细胞趋化蛋白-1(MCP-1)和脂联素蛋白分泌及mRNA表达的影响。方法体外培养SW872细胞,诱导细胞分化,不同浓度visfatin作用于SW872成熟脂肪细胞,孵育24h后收获细胞培养上清和RNA,分别采用ELISA法和RT-PCR法检测上清中TNF-α、MCP-1和脂联素的蛋白分泌及mRNA表达水平。结果与对照组比较,50μg/Lvisfatin即显著促进SW872脂肪细胞TNF-α、MCP-1分泌和mRNA表达及抑制脂联素分泌和mRNA表达;TNF-α、MCP-1和脂联素的蛋白分泌水平均较对照组显著增加[(169.77±13.69)vs(149.30±17.40)ng/g,(1.36±0.14)vs(1.21±1.09)μg/g,(258.53±58.59)vs(330.59±65.00)μg/gPa<0.05],TNF-α和MCP-1mRNA的表达较对照组显著增加[(0.50±0.02)vs(0.45±0.02),(0.55±0.03)vs(0.48±0.01)Pa<0.05],而脂联素mRNA表达较对照组显著下降[(0.90±0.02)vs(0.94±0.04)P<0.05]。当visfatin水平增加至250μg/L时,脂肪细胞TNF-α[(303.20±17.50)ng/g]和MCP-1[(2.00±1.27)μg/g]的蛋白分泌水平较对照组均显著上升(Pa<0.01),而脂联素的蛋白分泌水平[(151.59±36.38)μg/g]则显著减少(P<0.01);同时,TNF-α和MCP-1的mRNA表达水平[(0.82±0.06)ng/g,(0.87±0.06)μg/g]较对照组亦显著增加(Pa<0.01),而脂联素的mRNA表达水平(0.58±0.04)μg/g则显著减少(Pa<0.01)。结论visfatin通过调节促炎性因子和抗炎因子的表达参与了SW872脂肪细胞炎性反应的发生发展。 Objective To evaluate the effects of visfatin on the protein and mRNA expressions of inflammatory factors including tumor necrosis factor( TNF - α) , monocyte chemotactic protein 1 ( MCP - 1 ) and adiponectin in SW872 adipocytes. Methods Preadipocytes of the line SW872 were cultured and induced to differentiate. Visfatin of the doses 0 - 250 μg/L was added into the cultured fluid of the mature SW872 adipocytes for 24 h. ELISA and RT - PCR were used to detect the protein secretion and the mRNA expression of TNF - α, MCP - 1 and adiponectin. Results Compared with the control group (0 μg / L visfatin group) ,50 μg/L visfatin could significantly promote SW872 adipocytes TNF - α and MCP - 1 secretions and mRNA expression levels, as well as inhibiting the secretion and mRNA expression levels of adiponectin. MCP - 1 and adiponectin increased significantly than those in control group [ ( 169.77 ± 13.69) vs ( 149.30 ± 17.40) ng/g, (1.36±0.14) vs (1.21 ±1.09) μg/g,(258.53 ±58.59) vs (330.59 ±65.00)μg/g Pa 〈0.05].The expression of TNF-α and MCP-1 mRNA increased significantly than those in control group[(0.50 ±0.02) vs (0.45±0.02),(0.55±0.03) vs (0.48±0.01) Pa〈 0.05 ]. But the expression of adiponeetin was decreased significantly than that in control group [ (0.90 ± 0.02 ) vs ( 0. 94 ± 0.04 ) P 〈 0.05 ]. When the concentration of visfatin was increased to 250 μg/L,the fat cells TNF - α and MCP - 1 secretions of the protein levels were increasedsignificantly[(303.20±17.50) vs (149.30±17.40) ng/g P〈0.01,(2.00 ± 1.27) vs (1.21 ±1.09) μg/g P〈0.01], and protein secretion of adiponectin levels was decreased significantly [ ( 151.59 ± 36.38 ) vs (330.59± 65.00 ) μg/g P 〈 0.01 ] ; at the same time, TNF - αand MCP - 1 expressions of the mRNA level were increased significantly [ (0.82 ± 0.06 ) vs ( 0. 45 ± 0.02 ) , ( 0.87 ±0. 06) vs (0.48 ± 0.01 ) P 〈 0.01 ], while the mRNA expression of adiponectin was reduced significantly [ (0.58 ± 0.04 ) vs (0.94 ± 0.04) P 〈 0.01 ). Conclusions Visfatin may contribute to the development of the inflammation on SW872 adipocytes by regulating the pro - inflammatory factor and anti - inflammatory factor expressions.
