摘要
目的:评价增免升白冲剂的药效,探讨升高白细胞作用机理。方法:腹腔注射CP和^(60)Co照射LACA小鼠复制WBC减少模型,并经口给予不同剂量的该药治疗9d后计数WBC、BMC、CFU-S、CFU-GM并测定BMC中DNA含量。结果:1500mg·kg^(-1)治疗9d CP和^(60)Co模型小鼠WBC计数均恢复至正常,CP模型小鼠CFU-S和DNA含量亦恢复正常;3000mg·kg^(-1)治疗9d,两种模型小鼠各项检测指标均恢复至正常水平,疗效明显优于鲨肝醇;21例白细胞减少症志愿受试者按100mg·kg-1日剂量连续治疗30d,其中20例WBC计数均升至4×10~9·L^(-1)以上。结论:增免升白冲剂有明显的升高WBC、BMC、CFU-S、CFU-GM计数和增加DNA含量的作用,其疗效明显优于鲨肝醇,其机理可能是组方中各有效成分有改善造血微环境,促进DNA合成,促进造血干细胞增殖、分化和成熟作用。
OBJECTIVE: To study the mechanism and evaluate the medicative effects of IIRWP. METHODS: LACA mice model with leukopenia was made by administrating CP Pi and exposing 60Co - γ ray. After therapies with 3 doses IIRWP by gavage for 9 days, WBC,BMC,CFU - S,CFU - GM number and quantity of DNA in BMC was determined. RESULTS: When the CP and 60Co model LACA mice were treatd with 1500mg·kg-1 for 9 days, WBC number rose to normal level.SO were CFU - S and DNA quantity of mice. At 3000mg.kg-1 ,not only all indexes rose to normal levels,but also therapeutic effects of IIRWP were superior to those of batyl alcohol. 21 volunteer suffered leukopenia were treated continuously for 30 days with 100mg·kg-1 IIRWP, WBC number of 20 patients rose to exceed 4 × 109·?L-1. CONCLUSION:The IIRWP raise significantly WBC, BMC, CFU - S, CFU - GM, DNA and therapeutic effects were distinctly superior to those of batyl alcohol. The mechanism may be that effective components of every Chinese drug in IIRWP mixture can promote to synthesize DNA,accelerate proliferation and improve blood minute environment.
