甘草酸对小鼠安替比林及醋氨酚代谢的影响 被引量:9

摘要 目的研究甘草酸是否诱导小鼠肝细胞增殖及其对安替比林和醋氨酚代谢的影响。方法给小鼠甘草酸50mg·kg-1或等量注射用水灌胃7d后,比较两者间安替比林和醋氨酚有关药代动力学参数及肝DNA含量和肝/体重比的差异。结果与对照组相比,甘草酸组小鼠安替比林T12缩短453%,CL增加436%(P<005);醋氨酚T12延长546%,CL降低293%(P<005),尿液葡萄糖醛酸醋氨酚含量减少118%(P<005);肝DNA含量及肝/体重比无明显改变。结论甘草酸能诱导小鼠安替比林代谢,抑制醋氨酚代谢。 AIM To study the effect of pretreatment with glycyrrhizin on the hepatocyte proliferation and disposition of antipyrine and acetaminophen in mice. METHODS After administering ig 0 2% glycyrrhizin 50 mg·kg -1 or equal amount of distilled water to mice for 7 d, the changes of hepatic DNA content, ratio of liver/body weight and pharmacokinetic parameters of antipyrine and acetaminophen were investigated in a parallel controlled design. RESULTS In comparison with mice as control, T 12 of antipyrine was shortened by 45 3% and CL increased by 43 6% ( P <0 05) in mice pretreated with glycyrrhizn; while T 12 of acetaminophen was prolonged 54 6%, CL decreased 29 3% ( P <0 05), and the excretion of urinary glucuronide acetaminophen reduced by 11 8% ( P <0 05). The hepatic DNA content and ratio of liver/body weight insignificantly differed between mice with and without glycyrrhizin. CONCLUSION The ingestion of glycyrrhizin in mice induced the metabolism of antipyrine, inhibited acetaminophen disposition, but did not stimulate the hepatocyte prliferation.
