摘要
实验通过优化反相高效液相色谱法固相萃取和色谱分析过程中的检测参数,分析检测三角帆蚌体内MC—LR的含量。样品采用80%甲醇一水和丙酮混合溶剂提取,经ODSC18固相萃取柱萃取,将MC-LR从蚌器官内有效分离出来。以水(1‰TFA):甲醇=35:65(v/v)作为流动相,在238nm紫外波长下进行分析,绝对检测限为2.0ng,藻毒素浓度在0-25mg/L范围内,进样量与峰面积呈良好的线性关系,各组织器官平均回收率均达到70%以上。
Microcystins-LR in the Hyriopsis cumingii samples were determined by HPLC with solid phase extraction. An ODS C18 solid phase column extractor was used to extract the MC-LR from dif- ferent organs of mussel, and the muscle sample was extracted with a mixture of methanol-water (80%) and acetone. With a mobile phase of water (containing 1% trifluoroacetic acid):methanol =35:65(V/V), the ultraviolet wavelength was 238 rim. The method is sensitive with detection limits of 2.0ng, the lin- ear relationship between peak area and concentration of microcystin holds in the range of 0-25mg/L, and the average recoveries in four organs were over 70%.
