白细胞介素1对体外培养小鼠骨髓基质细胞增殖及细胞周期的影响

摘要 目的研究白细胞介素1对小鼠骨髓基质细胞增殖的影响。方法取出小鼠骨髓基质细胞作原代培养,用[3H]TdR参入法及MTT法观察其增殖情况,利用流式细胞仪观察其细胞周期的变化。结果白细胞介素1在250×103~1000×103UL-1范围内可促进小鼠骨髓基质细胞的增殖,并引起S期细胞的增多。结论白细胞介素1可促进体外培养的小鼠骨髓基质细胞增殖。 AIM To study the effect of interleukin1 on the growth of murine bone marrow stromal cells. METHODS Murine bone marrow stromal cells were taken out and incubated. The growth of the cultured cells were measured by TdR incorporation and MTT method, and the cell cycle of these cells were analysed by flowcytometry. RESULTS Interleukin 1(250×103 ̄1 000×103 U·L-1) prompted the proliferation of bone marrow stromal cells compared with the control group, induced anincrease in S phase cells of cell cycle detected by flowcytometry. CONCLUSION Interleukin 1 prompted the proliferation of murine bone marrow stromal cells, and induced increase in S phase cell of cell cycle.
