缝隙连接阻断剂-辛醇对实验性局灶性脑缺血/再灌注半暗带的影响 被引量:1

摘要 目的研究Wistar大鼠局灶性脑缺血/再灌注后72 h内不同时间点半暗带面积的变化及缝隙连接(GJ)阻断剂辛醇(Octanol)对半暗带面积的影响。方法雄性Wistar大鼠随机分为假手术组、对照组、辛醇干预组、DMSO溶剂对照组。大脑中动脉线栓法制备局灶性脑缺血/再灌注模型,Zea Longa评分观察行为学变化,免疫组化测定缝隙连接蛋白43(connexin43,Cx43)表达,甲苯胺蓝染色、焦油紫染色并利用图像分析仪测量半暗带面积。结果各组动物神经功能缺损评分无显著差异;缺血/再灌注后Cx43表达无明显变化;缺血/再灌注30 min开始半暗带面积逐渐扩大,于再灌注6 h达最大,以后逐渐缩小;辛醇干预组半暗带面积较对照组相比明显缩小,坏死神经元数目明显减少。结论脑缺血/再灌注对Cx43的表达无明显影响;缝隙连接阻断剂辛醇可明显缩小局灶性脑缺血/再灌注半暗带面积。 Objective To explore effect of gap junction blocker-octanol on penumbra at different ischemia/reperfusion periods within 72 h following cerebral ischemia/reperfusion in rats.Methods Wistar rats were randomly divided into sham-operated group,control group,octanol group and DMSO group.Rats underwent middle cerebral artery occlusion followed reperfusion with suture method.Behavior were evaluated by Zea Longa method.The brains were stained with toluidine blue and cresyl viollet.The expression of Cx43 was used by immunohistochemistry.The area of penumbra was analyzed with an image analysis system.Results There was no difference among groups except sham-operated about the results of behavior and Cx43 expression.The two methods correlated with each other in measuring penumbra.The area of penumbra expanded rapidly in a short period after reperfusion and peaked at 6 hours.Octanol could effectively decrease penumbra and neuron death.Conclusion Ischemia and reperfusion can not affect the Cx43 levels.The gap junction blocker-Octanol,can effectively decrease area of penumbra following MCAO in experimental cerebral ischemia.
