摘要
目的探讨七氟醚预处理对大鼠局灶性脑缺血再灌注损伤GFAP表达变化。方法建立大鼠七氟醚预处理及局灶脑缺血(PMCA)模型,应用免疫荧光方法观察七氟醚预处理(2%sevoflurane,2 h)+再灌注损伤后不同时间点(6 h、24 h4、8 h、72 h)GFAP表达的变化。结果再灌注损伤后随时间延长,GFAP表达逐渐增多,于缺血后24 h达高峰。七氟醚预处理各时间点GFAP表达明显降低,尤其在缺血周边区明显,与缺血对照组相比差异具有显著性(P<0.05)。结论七氟醚预处理明显抑制星形胶质细胞活化及反应性增生可能是七氟醚预处理的脑保护机制之一。
Objective To study the expression change of GFAP after sevoflurane preconditioning and focal cerebral ischemia.Methods The sevoflurane preconditioning model and PMCAO model were made and the expression of GFAP was examined by immunofluorescence staining at 6 h、24 h、48 h、72 h after cerebral ischemia/reperfusion.Results After reperfusion,the expression of GFAP gradually increased and peaked at 24 h after ischemia.In the isoflurane preconditioning 2 h +focal cerebral ischemia group,the expression of GFAP increased significantly in the infarct border compared with ischemia control group(PMCAO)(P〈0.05).Conclusion The activation and proliferation of astrocytes was significantly suppressed by sevoflurane-preconditioning,which might be one of the protective mechanism of focal cerebral ischemia-reperfusion injury.
