急性病毒感染对小鼠心肌糖蛋白130的基因及蛋白表达的影响

摘要 目的:探讨糖蛋白130(glycoprotein 130)在病毒性心肌炎中的作用。方法:建立病毒性心肌炎小鼠模型,观察其心肌病理和血清IL-6水平;用RT-PCR方法观察小鼠心肌gp130 mRNA的表达;Western blot方法观察gp130蛋白的表达。结果:病毒组与正常对照组相比,病毒组小鼠心肌病变检出率和血清IL-6水平显著高于对照组(P<0.001);心肌gp130 mRNA表达亦见明显增高(P<0.01),其蛋白表达未见明显改变(P>0.05)。结论:急性病毒感染促使小鼠心肌gp130基因表达,机制有待进一研究。 Objective: To study the role of glycoprotein (gp)130 in myocarditis. Methods:Thirty-two BALB/c mice were randomly divided into 2 groups, i.e. Coxsakievirus B3 (CVB3)infection group and control group. The myocardial injury of the heart biopsy samples and serum IL-6 levels were examined. The expression of gp130 mRNA and protein were observed by RT PCR and western blot, respectively. Results: Compared with those of controls, the rate of incidence and the extent of myocar- dium injury and serum /L-6 level of infected mice were significantly increased(P〈0. 001 ), and the expression of gp130 mRNA were also increased(P〈0.01 ), but the expression of gp130 protein were not changed significantly(P〉0.05). Conclusion:CVB infection would increase the expression of gp130 gene in mice myocardium and the mechanism of such up-regulation needs fur ther exploration.
