摘要
根据已发表的猪圆环病毒2型(PCV-2)的全基因组序列,设计1对特异性引物,进行PCR扩增,将1个扩增产物连接到pUCm-T vector上并克隆到大肠杆菌DH5α中,得到1个阳性克隆,经提取质粒进行PCR、酶切、测序鉴定,证明扩增出了PCV-2目的片段。结果表明本实验分离株(JXPCV-2)与国内外不同地域的PCV-2全基因组序列的差异很小,同源性在94.2%~99.7%;进化树分析表明JXPCV-2与其他10株PCV-2分离株的亲缘关系较近,与浙江分离株(AY691169)亲缘关系最近。
According to the published genome sequences of porcine cireovirus type 2 ( PCV - 2), a pair of special primer was designed to detect PCV -2 in diseased or dead swine specimens from different pig farms in Jiangxi by PCR. The PCR product was extracted and linked to pUCm -T vector and then cloned into E. coll. DH5α. The recombination plasmid was identified by digestion with different enzyme restrictions, PCR and se, queneing analysis on the the complete genome sequences, the JXPCV -2 isolate shared higher homology with the isolates from internal and overseas regions, which is from 94.2% - 99.7%. Phylogenetie tree analysis revealed that JXPCV -2 was closely related to the other ten isolates, and related most colsely to Zhengjiang isolate AY691169.
