Expression of heat shock proteins in peripheral blood lymphocytes and the significance in patients with non-small cell lung cancer

摘要 目的探讨非小细胞肺癌(NSCLC)患者外周血淋巴细胞热休克蛋白(HSP)27、HSP72和HSP73的表达水平及其与临床病理生理特征的关系。方法采用流式细胞术检测34例NSCLC患者外周血淋巴细胞HSP27、HSP72和HSP73的表达水平,并以40例健康受试者作为对照。结果NSCLC外周血淋巴细胞HSP27、HSP72的表达量均低于对照组(P〈0.05),两者表达量呈正相关(P〈0.01),而HSP73的表达量和对照组差异无统计学意义(P〉0.05)。NSCLC外周血淋巴细胞HSP27的表达量与临床病理生理特征无明显相关,而HSP72的表达量与NSCLC原发肿瘤的大小、淋巴结转移及临床分期相关(P〈0.05)。结论NSCLC患者外周血淋巴细胞HSP27、HSP72表达下调,且HSP72与NSCLC的发生发展及淋巴结转移相关。 Objective To investigate the relationship between the expression levels of HSP27, HSP72 and HSP73 in peripheral blood lymphocytes and clinical pathophysiological characteristics in patients with non-small cell lung cancer (NSCLC). Methods Flow cytometry was used to detect the expression of HSP27, HSP72, and HSP73 in peripheral blood lymphocytes of 34 patients with NSCLC and 40 healthy controls. Results The expression levels of HSP27, and HSP72 in peripheral blood lymphocytes of NSCLC patients were decreased as compared with those in normal controls ( P 〈 0.05 ). The correlation analysis showed that HSP27 level was positively correlated to HSP72 level (P 〈 0.01, r = 0.60). No statistical difference was observed in HSP73 between the two groups ( P 〉 0.05 ). The expression level of HSP27 in peripheral blood lymphocytes had no relation to clinicopathophysiological characteristics of patients with NSCLC. The expression level of HSP72 in peripheral blood lymphocytes was related to size of tumor,lymph node status and clinical stages ( P 〈 0.05 ), but no to sex, age and pathological types of NSCLC patients (P 〉 0.05). Conclusion The expression of HSP27, and HSP72 is decreased in peripheral blood lymphocytes of patients with NSCLC, and HSP72 may play an important role in tumor growth and lymphatic metastasis. The level of HSP72 in peripheral blood lymphocytes may be used as a predictor for prognosis.
