摘要
目的测定不同年龄组颈椎椎间盘中的蛋白多糖酶活性,探讨蛋白多糖酶活性与年龄之间的相关性。方法对40个不同年龄颈椎间盘的蛋白多糖酶活性进行了测定,分别来自胎儿组、17~25岁组、40~60岁组以及60~75岁组,用含有10.0mmol/LCaCl2的0.05mol/LTris/HC对椎间盘中的蛋白多糖酶进行提取,用1mmol/L APMA将其激活,与取自牛肩胛软骨的蛋白多糖单体底物进行反应,以酶标仪来测定蛋白多糖酶的活性。结果胎儿组、17~25岁组、40~55岁组和60~75岁组颈椎间盘的蛋白多糖酶活性的吸光度值分别是1.911±0.21、0.881±0.155、1.720±0.19、1.777±0.142。胎儿组颈椎间盘与60~75岁组颈椎间盘的吸光度值最高,两者之间差异无统计学意义(P〉0.05),胎儿组与17~25岁组、40~55岁组颈椎间盘的吸光度值结果差异有统计学意义(P〈0.05),17—25岁组的吸光度值最低,与胎儿组、40~55岁组和60~75岁组的测得值差异有统计学意义(P〈0.01),40~55岁组和60~75岁组的吸光度值差异无统计学意义(P〉0.05)。结论颈椎间盘中的蛋白多糖酶活性与年龄明显相关。
Objective Experimental study on age-related changes in activities of proteoglycanases obtained from various cervical intervertebral discs. Methods 40 spcimens were analysed, among these lumbar intervertebral discs, 10 were fetal cevical intervertcbral discs, 10 were 17-25 years-old cervical intervertebral discs, 10 were 40-55 years-old cervical intervertebral discs, 10 were 60-75 years-old cervical intervertebral discs. Proteoglycansaes were extracted with 0.05 mmol/L Tris/HCl buffer which contains 10.0 mmol/L CaCl2. The proteoglycan of bovine scapula cartilage extracted with 4.0mol/L Guanidine HCl was used as substrate. The proteoglycan breakdown of the reaction indicated the activity of the proteoglycanases from various luumbar intervertebral discs. Results Activities of proteoglycanases from fetal discs, 17-25 years-old discs,40-55 years-old discs and 60-75 years-old discs were 1. 911± 0. 21,0. 881 ±0. 155,1. 720 ± 0. 19,1. 777 ±0. 142. Conclusion Levels of Proteoglycausae were age-related and may play a important role in the pathological process of degeneration of cervical intervertebral discs.
