摘要
目的:探讨血管紧张素转换酶(ACE)在香烟诱导的仓鼠肺动脉高压(PAH)肺小动脉重构病变中的表达及对血管紧张素Ⅱ(AngⅡ)生成的作用。方法:将20只雄性仓鼠随机分为对照组和熏烟组。仓鼠暴露于香烟烟雾中建立肺动脉高压模型。采用免疫组织化学染色法检测肺组织中肺小动脉的ACE蛋白表达;采用酶联免疫吸附实验测定肺组织ACE含量;采用放射免疫分析法测定肺组织AngⅡ含量。结果:(1)ACE在熏烟组仓鼠肺组织中的表达明显增加。(2)熏烟组肺组织匀浆中AngⅡ和ACE含量均较对照组明显升高(P均<0.05)。结论:香烟暴露可刺激ACE在肺小动脉重构中的表达增加,并且ACE在香烟诱导的仓鼠肺小动脉重构病变中对AngⅡ生成起作用。
Objective: This study was to assess the expression of ACE in cigarette smoke-induced pulmonary arterial remodeling. Method: In this study, 20 hamsters were randomly divided into 2 groups: control and smoke exposure only. After four months smoke exposure, lung morphology was examined by Immunohis- tochemistry, and then tissue biochemical changes were measured by radioimmunoassay and ELISA. Results: The levels of lung tissue ACE increased in chronic cigarette smoke-induced remodeling of pulmonary arterioles in hamsters, The increase of ACE was related to increased formation of Ang Ⅱ. Conclusion: This study demonstrated that ACE up-regulated in smoke-induced pulmonary arterial remodeling and contribu- ted to the enzymatic production of AngⅡ.
