从红树林内生真菌筛选重组人DNA拓扑异构酶Ⅰ抑制剂的研究 被引量:1

Screening of inhibitors of recombinant human DNA topoisomerase Ⅰ from mangrove endophytic fungi

摘要 从6种红树林植物中分离到60株内生真菌。通过重组人DNA TopoisomeraseI(hTopoI)的松弛活性的抑制实验筛选发现:有45%的菌株代谢产物对hTopoI松弛质粒抑制活性的IC50小于1/2,并筛选到两株对hTopoI具有强烈抑制活性的内生真菌MF54和MF60,其IC50均为1/128。以上研究结果表明:一些红树林内生真菌的代谢产物对hTopoI表现出良好的抑制活性,有望从中发现潜在的抗肿瘤先导化合物。 Sixty strain endophytic fungi were isolated from six mangrove plants. The hTopo I-inhibiting ability of the extracts from 60 strains of mangrove fungal endophytes were investigated by inhibition assay of relaxing activity of hTopo 1. 45% of them had good activity to inhibit hTopo I (IC50〈1/2), and two cultures (MF54 and MF60) possessed higher activity of inhibit hTopo I (their IC50= 1/128), which will potentially become new antitumor drugs. The results indicated that some metabolites of mangrove fungal endophytes were potential resources of compounds of new antitumor activities, which are worthy of further research and development.
