摘要 高效TAIL-PCR是一种完全依赖PCR的方法,它利用5'端带尾巴的高简并倍数的LAD引物在未知序列上创造结合位点;LAD0引物是LAD系列简并引物5'端的部分序列,利用其可以保证PCR扩增中结合未知序列端的引物浓度;利用巢式PCR可提高扩增产物特异性。本研究将该技术应用于洋葱的未知侧翼序列分离,建立了洋葱的高效TAIL-PCR技术体系。 The hiTAIL - PCR is an effective method completely based on PCR technique. A long arbitrary degenerate (LAD) primer with a higher degree of degeneracy is used to create primer - binding sites efficiently along the unknown target sequences. An additional sequence LAD0 identical to the 5' part of the LAD primer is utilized, resulting in primer concentration that is high enough to bind the unknown flanking sequence. Using the nested PCR is intended to improve specificity of PCR products. We firstly established the hiTAIL- PCR system to isolate the unknown target sequences in onion.
