摘要 目的建立清开灵颗粒中黄芩苷和绿原酸的含量测定方法。方法色谱柱为KromasilC18(4.6mm×250mm,5μm),流动相为甲醇-水-磷酸(20∶80∶0.2),流速1ml/min,柱温25℃,检测波长327nm。结果黄芩苷在0.09~0.54μg之间线性关系良好,平均回收率100.3%,RSD=0.99%;绿原酸在0.196~1.176μg之间线性关系良好,平均回收率99.9%,RSD=1.06%。结论本法灵敏、简便、准确,可用于清开灵颗粒的测定和质量控制。 Objective To establish determination of scutellarin and chlorogenic acid in Qingkailing Extract by HPLC. Methods Kromasil C18 column and the mobile phase with methnol-water-phosphoric acid (20 : 80: 0. 2) with the flow rate 1.0 ml/min and column temperature 25℃ and the detection wavelength at 327 nm were applied for the determination. Results The linear range of scutellarin in the range of 0. 090 - 0. 540μg, r =0. 9999, and its average recovery rate 100. 3% ( RSD =0.99% ) ; while the linear of chlorogenic acid in the range of 0. 196 - 1. 176μg, r = 0. 9999, and its average recovery rate 99.9% ( RSD = 1.06% ). Conclusion The method is sensitive, simple, accurate, so HPLC can be used for the determination of prepara- tions and the quality control in Qingkailing Extract.
