造血干细胞衰老模型的建立及其评价 被引量:1

摘要 造血干细胞衰老模型的建立是研究造血干细胞衰老机理与延缓其衰老的重要途径,目前可通过体内和体外两种途径实现。体内衰老模型可通过供体造血干细胞在受体内连续性移植建立,体外衰老模型可通过白消安、三丁基过氧化氢等致衰老药物和电离辐射等方法建立。造血干细胞衰老模型建立后,可通过其自我更新和多向分化能力检测、细胞周期分析、β-半乳糖苷酶染色等方法检测和评价造血干细胞的衰老变化,并应用Southern bloting,Western bloting,RT-PCR等方法检测造血干细胞衰老相关基因和蛋白变化。本文就造血干细胞衰老模型的建立以及评价方法综述如下。 The establishment of hematopoietic stem cells aging model is an important way to study the mechanism of hematopoietic stem cells aging and to delay their aging, which can be achieved by in vivo and in vitro models. The in vivo model can be established through the hematopoietic stem cells serial transplantation from the donor to the receptor. The model in vitro can be set up using the ionizing radiation and the medicine to induce cell aging, such as busulfan and tertbutylhydroperoxid. After the model established, the aging changes of hematopoietic stem cells can be detected and evaluated through the capability of self-renewal and the multi-differentiation, cell cycle analysis and β -Gal staining. The changes of the hematopoietic stem cells senescence-associated genes and proteins can be detected through the Southern hloting, Western bloting and RT - PCR. The review will summarize the establishment of the aging model and its evaluation methods.
