摘要
为了研究弓形虫代谢分泌抗原对仔猪T细胞亚群及抗体的影响,将仔猪随机分为6组,每组5只,分别为E/SA组、E/SA+CPG(未乳化)组、E/SA+CPG组、E/SA+IL-2组、E/SA+IL-2+CPG组及对照组。分别于免疫前、免疫后第2周、免疫后第4周、感染后第1周及感染后第2周采集仔猪外周血,用流式细胞仪对各试验组外周血中CD4+及CD8+T淋巴细胞亚群的动态变化规律进行检测,用IHA对抗弓形虫抗体水平做检测。结果显示,弓形虫代谢分泌抗原免疫仔猪后第4周,免疫组外周血CD4+、CD8+T淋巴细胞水平和特异性抗体效价与对照组相比均有大幅度升高;感染后第1周,免疫组CD4+T细胞水平及CD4+/CD8+比值较感染前有不同程度降低,感染后第2周,免疫组CD4+T细胞水平及CD4+/CD8+比值上升至免疫后水平,CD8+水平下降,感染后特异性抗体效价进一步升高。表明,弓形虫代谢分泌抗原能够提高仔猪外周血CD4+及CD8+T淋巴细胞水平和特异性抗体水平。
To elucidate the effects of ToxopIasma gondii excreted/secreted antigen(E/SA) on piglet's T lymphocyte subgroups and antibodies, piglets were randomly divided into six groups of E/SA, E/SA+CpG(unemulsification) ,E/SA + CpG, E/SA + IL-2, E/SA + IL-2 + CpG groups and the control group. The dynamic changes of the CD4^+ and CD8^+ T lymphocyte subgroups in the piglet' s peripheral blood were determined by flow cytometry, and antibody levels were tested by indirect hemagglutination assay. The results showed that on weeks four post-immunization with T. gondii E/SA, the peripheral blood CD4^+ and CD8^+T lymphocyte levels and specific antibody levels of the immune groups had significantly increased compared with those of the control group. The specific antibody levels further increased on week 1 after inoculation,the CD4^+ T cells levels and CD4^+/CD8^+ ratio of the immune groups reduced to varying degrees than two weeks post infection, CD4^+ T cells levels and CD4^+/ CD8^+ ratio in the immune group increased to immunized levels,CD8^+ levels declined. So the T. gondii E/SA can improve the percentage of the piglet's CD4^+ and CD8^+ T lymphocytes and the specific antibody levels in peripheral blood.
