摘要
目的应用基因芯片技术探讨外周血单个核细胞(PBMC)及心房组织细胞因子的差异表达基因,探讨不同组织对体外循环(CPB)的反应。方法CPB开始、CPB结束即刻抽取患者动脉血,密度梯度离心法分离PBMC;取CPB开始及CPB结束即刻的右房组织。用BD AtlasTMcDNA Expression Arrays表达谱基因芯片对比CPB前及CPB后二者间细胞因子的差异表达。结果CPB前PBMC及心房间细胞因子基因表达差异明显者有IL-6、IL-13、W nt5 a、促肾上腺皮质激素释放激素Ⅰ型受体、卵泡刺激素受体和盘状结构域受体2。CPB后二者的基因表达谱均发生变化,差异明显者变为IL-6、IL-13、α-干扰素诱导蛋白(IFI616)、钙结合蛋白S100A9(钙颗粒素B)、胎盘生长因子和W nt5 a。结论CPB前PBMC与心房组织的细胞因子基因表达谱不同,CPB刺激后基因表达谱发生变化,但变化不一致,使差异表达基因发生变化。
OBJECTIVE To determine the differentially expressed genes of cytokines between peripheral blood mononuclear cell (PBMC) and fight atrium using gene chip technique. To explore the reactions to eardiopulmonary bypass (CPB) in the different tissues. METHODS The patients' oxygenated bloods were drawn immediately before onset and termination of CPB. PBMC were obtained from heparinised blood by Ficoll gradient centrifugation. Right atrium were col- lected just before onset and termination of CPB. The differential expression between PBMC and atrium was compared using BD AtlasTM cDNA Expression Arrays. RESULTS Between PBMC and fight atrium before CPB, the differentially expressed genes were interleukin 6, intedeukin 13, Wnt5a, corticotropin releasing hormone receptor 1 (CRH1R), follicle stimulating hormone receptor ( FSH - R) and discoidin domain receptor family, member 2 ( discoidin domain receptor, DDR2). Both of the gene expression profiles changed after CPB. The differentially expressed genes between PBMC and right atrium were interleukin 6, interleukin 13, interferon alpha- inducible protein (clone IFI -6 -16), S100 calcium binding protein A9 (ealgranulin B) , placental growth factor and WntSa. CONCLUSION The gene expression profiles in cytokines of PBMC and right atrium were different before CPB. The gene expression profiles changed in different manners after CPB. So, the differentially expressed genes changed.
