摘要
目的观察重症胸部创伤大鼠Th1/Th2细胞平衡的变化,评价匹多莫德对重症胸部创伤大鼠Th1/Th2细胞平衡的影响。方法20只雄性SD大鼠随机平均分配到单纯创伤组(A组)和创伤+匹多莫德组(B组)。大鼠均经胸部撞击致伤建立重症胸部创伤模型,B组大鼠每日予匹多莫德100mg/kg腹腔注射,A组大鼠予等体积生理盐水腹腔注射。分别于受伤前及受伤后第1、3、5、7天采取全血各1.0ml,分离外周血单个核细胞(PBMC),体外培养72h,收集上清液冻存,ELISA法集中检测IFN-γ、IL-4浓度。结果两组大鼠PBMC培养上清液中IFN-γ浓度在受伤后均迅速升高,随即回落,B组明显高于A组,差异有统计学意义(P<0.05);IL-4浓度在受伤后均持续升高,两组间差异无统计学意义(P>0.05);IFN-γ浓度上升倍数/IL-4浓度上升倍数比值在受伤后均升高,随后逐步降低至1.0以下,B组高于A组,差异有统计学意义(P<0.05)。结论大鼠在重症创伤后早期,Th1细胞功能迅速增强,随后逐步受到抑制,Th2细胞功能则持续增强,发生Th1细胞向Th2细胞的漂移。匹多莫德提高了重症创伤大鼠IFN-γ的合成分泌,减轻了Th1细胞功能抑制的程度,有助于维持Th1/Th2细胞平衡。
Objective To observe the Th1/Th2 equilibrium in rats with severe thoracic injury,and evaluate effect of pidotimod on the equilibrium. Methods Twenty male Sprague-Dawley rats were ran- domized into group A and group B.All the animals were subjected to impact on the right lateral superior chest. Pidotimod was administered to rats in group B,by intraperitoneal route (100 mg·kg-1 ·d-1). Periph- eral blood mononnclear cells(PBMCs) were harvested prior to injury,and the 1st, 3rd, 5th, 7th day af- ter injury, then cultured for 72 hours. Concentrations of IFN-γ and IL-4 in the supernatant were mea- sured by means of enzyme linked immunosorbent assay. Results Post-injury concentration of IFN-γ in PBMC supernatant increased significantly in both groups, then decreased gradually. The concentration of IFN-γ in group B was higher than that in group A (P〈0.05). Concentration of IL-4 in PBMC supematant also increased significantly in both groups, and concentration of IL-4 in group B was similar to that in group A. The "△IFN-γ/AIL-4"(multiplying power of IFN-γ/ multiplying power of IL-4) increased in both groups, then decreased gradually and fall into 1.0. The "△IFN-γ/AIL-4" in group B was higher than that in group A(P〈0.05). Conclusion Suffered from severe thoracic injury, the Thl and Th2 cell function of the rats enhances together in the early post-injury period.The Thl cell function is suppressed soon,but the Th2 cell function enhances constantly. By up-regulating IFN-γ production, pidotimod dimin- ished the suppression of Thl. Pidotimod can help maintain the Thl/Th2 equilibrium in the severe tho- racic injury rats.
