摘要
目的探讨 Caspase-3在肝癌发生发展中的作用和意义。方法应用免疫组织化学SP法,对二乙基亚硝胺(DEN)诱发大鼠肝癌发生过程中 Caspase-3的表达进行了系统观察。结果①DEN诱发大鼠肝癌发生率为100%;②在正常大鼠肝脏、诱癌早期(1—8周)及诱癌中期(9—16周)可见少量肝细胞及枯否细胞呈 Caspase-3阳性表达,随着肝癌发生进程, Caspase-3阳性枯否细胞数量增多,在癌结节中可见 Caspase-3阳性肝细胞,并且结节周边部比中央部阳性细胞多。结论本研究表明肝细胞 Caspase-3的低表达和枯否细胞 Caspase-3的高表达可能与肝癌的发生和发展有关。
Objective To investigate the expression of Caspase-3 during hepatocarcinogenesis induced by diethylnitrosamine (DEN) in rats systematically. Methods The expression of Caspase-3 was measured by immunohistochemical SP method. Results (1) The incidence of hepatocarcinoma induced by DEN was 100%. (2)The expression of Caspase-3 was detected occasionally in hepatocytes and Kupffer cells of normal rats and hepatoma rats with DEN treatment for 1-16 weeks. With the progress of hepatocarcinogenesis , the number of kupffer cells expressing Caspase-3 increased progressively,and the number of hepatocyes expressing Caspase-3 in periphery of hyperplastic nodule was more than that in the centre. Conclusion The results suggest that the low expression of Caspase-3 in hepatocytes and the high expression of Caspase-3 are related closely to the development and progression of hepatoma.
