白细胞介素-3在变应性鼻炎体质模型鼻腔黏膜中的表达及意义

Expression and role of nasal mucous interleukin-3 in constitutional models of allergic rhinitis

目的研究不同体质模型中变应性鼻炎的鼻腔黏膜白细胞介素-3(IL-3)的表达及意义。方法采用寒、热性中药及0.9%氯化钠注射液灌饲及同类传代的方法构建不同的体质模型;在此基础上采用卵清蛋白腹腔注射及滴鼻的方法构建变应性鼻炎的疾病模型,瑞氏染色计数鼻黏膜组织的嗜酸性粒细胞和肥大细胞,免疫组化检测鼻腔黏膜IL-3的表达。结果寒、热、常体的变应性鼻炎体质模型建立后,寒体组症状最为严重,热体组症状最轻微。鼻腔黏膜IL-3在各组体质的变应性鼻炎中均能表达,且寒体组表达最明显(P<0.05),并与鼻黏膜组织中的嗜酸性粒细胞和肥大细胞呈正相关(P<0.05)或显著正相关(P<0.01)。结论体质在变应性鼻炎的免疫调节中起关键作用,能够影响IL-3的表达。IL-3募集、趋化了嗜酸性粒细胞和肥大细胞在鼻腔黏膜的浸润和表达,能够反映变应性鼻炎的严重程度。

Objective To determine the expression of nasal mucous IL-3 in different constitutions of allergic rhinitis by experiment.Methods The different constitutional models were established by given cold,warm Chinese herbal medicine and normal saline.On the basis,the allergic rhinitis models were constructed by intraperitoneal injection and nasal drip with ovalbumin（OVA）.The eosinophils and mast cells in nasal mucous were counted Wright stain,and the expression of IL-3 in nasal mucous was detected by immunohistochemistry.Results The stable constitutional models of allergic rhinitis with cold,warm and normal were successful.The symptoms of the cold constitution are more serious than the warm constitution（P〈0.05）.IL-3 in nasal mucous can be expressed in all constitutions of allergic rhinitis and the cold constitution was the most distinct（P〈0.05）.Correlative analysis showed that there was a positive correlation（P〈0.05） or significant positive correlation（P〈0.01） between IL-3 and eosinophils,mast cells in nasal mucous.Conclusion Constitution plays a key role in immunoregulation of allergic rhinitis,which determined the expression of IL-3.IL-3 was an important cytokine to recruit eosinophils and mast cells in nasal mucous and reflect the severity of inflammation.
