摘要
目的优选小儿热清按摩凝胶中透皮促进剂氮酮的加入比例,研究黄芩苷的体外透皮吸收情况。方法采用Franz小池扩散法,以离体小鼠皮肤为透皮屏障,50%乙醇为接收介质,用HPLC法测定接收液中黄芩苷的含量,计算累积渗透量Q,拟合体外透皮释药方程,计算渗透速率,以此评价黄芩苷透过小鼠皮肤的能力,并筛选透皮促进剂的最佳比例。结果当氮酮浓度为2%时,黄芩苷6 h体外累积渗透量最高3.940 mg/cm2,渗透速率最大0.629 65 mg/(cm2.h),促透效果最佳。结论氮酮的加入可以明显促进黄芩苷的透皮吸收作用,但应控制适宜的浓度,本凝胶加入2%的氮酮促透作用最好。
Objective To select the optimum ratio of azone(transdermal accelerator) in Xiao'er're'qing massage gel and to study percutaneous absorption of baicalin in vitro.Methods The release rate of baicalin from the gel was detected by modified Franz diffusion cells method.Mouse skin was used as permeation barrier and 50% alcohol solution was used as release media.The content of baicalin in release media was measured by HPLC.The percutaneous absorption ability of baicalin was evaluated and the optimum ratio of transdermal accelerator was optimized by accumulative osmolality and permeate velocity with percutaneous absorption equation.Results The accumulative osmolality for 6h(3.940mgocm-2) and the permeate velocity(0.62965 mgocm-2oh-1) of baicalin in vitro was the best when the ratio of azone was 2%.Conclusion The adding of azone can obviously improve percutaneous action,but the content of azone should be controlled.The penetration-promoting action was the best when 2% azone was added in Xiao'er're'qing massage gel.
