重组血红素加氧酶-1乳酸乳球菌灌胃后大鼠肠道中血红素加氧酶-1重组蛋白含量变化 被引量:2

摘要 目的观察重组血红素加氧酶-1(heme oxygenase-1,HO-1)乳酸乳球菌灌胃后大鼠肠道中重组HO-1蛋白的含量。方法制备HO-1乳酸乳球菌悬液和乳酸乳球菌悬液,不同剂量和不同次数灌胃SD大鼠,在规定时间取回肠组织,酶联免疫吸附法和免疫组化法分析回肠组织HO-1的含量和分布情况。结果重组HO-1蛋白吸收后进入肠上皮细胞的胞膜和胞质中。回肠组织中HO-1含量:5倍、10倍HO-1乳酸乳球菌灌胃比20倍HO-1乳酸乳球菌灌胃高(P<0.01),连续灌胃4天、7天比连续灌胃10天含量高(P<0.01)。结论乳酸乳球菌中重组HO-1蛋白可以被回肠吸收,但在一定程度后增加HO-1乳酸乳球菌灌胃剂量和次数不能有效增加回肠中重组HO-1蛋白的含量。 Objective To investigate the variations in the intestinal content of recombinant heme oxygenase-1(HO-1) following gastrogavage of the HO-1-secreting Lactococcus lactis strain.Methods The HO-1-secreting Lactococcus lactis strain and Lactococcus lactis suspension were prepared by culture and centrifugation.The healthy SD rats underwent gavage of Lactococcus lactis strain at different doses and different time points.Immunohistochemistry and ELISA method were employed to determine the distribution and HO-1 content in ileal tissue.Results The Absorbed recombinant HO-1 was mainly located in the cytoplasm and cytomembrane of ileal tissue.The HO-1 content in the ileal tissue in low(5 and 10 times) dose groups was higher than that in high(20 times) dose groups(P〈0.01);the HO-1 content in ileal tissue in short gavage duration groups was higher than that in long(10 days) gavage duration groups(P〈0.01).Conclusion While HO-1 can be Absorbed by the ileum,the increase in gavage doses and times of the HO-1-secreting Lactococcus lactis cannot effectively increase the content of HO-1 in the ileum.
