摘要
利用PK15细胞从猪断奶后多系统衰竭综合征(PMWS)临床发病与死亡猪中分离到2株病毒;分别命名为Haian(登录号为FJ712216)和Xuancheng(登录号为FJ712215)。利用间接免疫荧光(IFA)和聚合酶链式反应(PCR)对其进行鉴定,并对其全基因组序列进行了测定和分析。结果表明,分离株感染细胞后病毒抗原主要分布在细胞核中,2分离株的基因组全长为1767nt,与世界上其他地区的PCV2分离株密切相关,核苷酸序列同源性达93.7%-97.7%,属于毒力较强的Genotype1。
Two porcine cireovirus type 2 (PCV2) strains, named Haian (FJ712216) and Xuancheng( FJ712215 ), were isolated from lymphoid node samples of clinically postweaning mnhisystemic wasting syndrome (PMWS) infected and dead pigs by PK15 cell culture. The results showed the two strains of PCV2 distributed mainly in the infected cell nucleolus by indirect immunofluorescence assay (IFA) ; The complete genomes of the two isolated strains were sequenced and ana- lyzed. The results showed that their genomes were both 1 767 nt in length. The sequences are closely related to those of other PCV2 isolates in the GenBank ,and the homology was 93.7% -97.7%. It was presumed that the two isolated strains were high pathogenic PCV2 genotype 1.
