日本血吸虫抱雌沟蛋白筛选噬菌体展示抗原对小鼠血吸虫病免疫效果观察 被引量:3

Immunological Effects of Schistosoma japonicum Gynecophoral Canal Protein on Mice with Phage-displayed Schistosoma

摘要 在前期研究中我们利用日本血吸虫抱雌沟蛋白筛选日本血吸虫噬菌体展示cDNA文库,共获得10种阳性噬菌体克隆。本研究选择其中5种噬菌体克隆单独或联合免疫小鼠,观察其对血吸虫成虫合抱的影响以及对血吸虫病的免疫预防效果。结果显示,19号克隆单独免疫和6号、18号混合免疫在预防实验中分别取得了40.53%和31.14%的减虫率及35.31%,44.65%的肝脏减卵率。研究结果提示,19号、6号和18号噬菌体展示的EST序列编码基因具有一定的抗血吸虫生殖免疫效果,在血吸虫疫苗研究方面具有重要价值。 In preliminary studies,10 positive phage display clones were screened by Schistosoma japonicum gynecophoral canal protein as Schistosoma japonicum phage display library.In this study,5 kinds of mice with independent and mixture immunity were selected to observe the effects of Schistosom disease on them.The result showed that 40.53% and 31.14%(clone 19) and 35.31% and 44.65%(clone 6 and 8) of mean adult worm burdens and liver egg burdens,respectively were obtained by the experiment of No.19 clone independent immunity and No. 6,No. 18 mixture immunity. The final results prompted that the EST sequence code gene in No. 19 ,No. 6 and No. 18 phage-displayed performed the significant immunity effect.
