摘要
利用ISSR分子标记技术对细叶桉的遗传多样进行了分析.从100条随机引物中筛选出10条特异的用于细叶桉ISSR-PCR扩增引物,扩增出101条清晰可重复的条带,其中89条是多态条带,多态带百分率为86.1%.采用POPGENE软件,计算了种间的Nei氏遗传距离,并用UPGMA法构建了聚类分析树形图,分析了它们的遗传多样性和亲缘关系.
The genetic diversity of Eucalyptus tereticornis was analyzed by ISSR technology.According to the improved ISSR-PCR system,10 primers were selected out of 100 random ones to yield specific bands with moderate band numbers,clear background and good reproducibility.Totally,101 specific and reproducible DNA bands were obtained,of which 89 are polymorphic bands(86.1% of polymorphism).Bio-software POPGENE 1.31 and NTSYS-pc were used to calculate the Nei genetic distance and produce the UPGMA dendrogram of genetic diversity and similarity.
