摘要
采用常规医用抗生素防治大多数细菌感染的方法,针对邓恩桉组织培养过程中试管苗的细菌污染问题,分别用注射器在分化培养基和生根培养基中加入不同浓度的不同抗生素.结果表明:培养基附加氨苄西林钠100 m g/L,对当代污染菌的抑菌率为100%,效果最好,同时相对在增殖系数、苗高、生长势方面都表现最好,并对邓恩桉试管苗的生根有明显的促进作用,可消除根的黄化现象.硫酸链霉素抗菌效果次之但对试管苗产生毒害,庆大霉素效果抗菌不佳.
Antibiotics were applied to control bacteria in the trail.In order to solve the problem of contamination during the process of tissue culture of E.dunnii,different concentrations of different antibiotics were added to subpassge medium and rooting medium separately.The results showed that medium adding with ampicillin sodium 100 mg/L was most effective: the inhibition rate of bacteria contamination for the contemporary generation was 100%;and at the same time proliferation coefficient,height and growth potential of the in vitro seedlings were performed the best;in additional,the rooting of in vitro seedlings were accelerated and etiolation phenomena of the roots was eliminated.The antibacterial ability of streptomycin sulfate was the second best but streptomycin sulfate will poison the in vitro seedlings,and the antibacterial ability of gentamicin was underperformed.
