N-乙酰半胱氨酸对波动性糖环境下脐静脉内皮细胞损伤的保护作用 被引量:1

摘要 目的:研究N-乙酰半胱氨酸(NAC)在高糖及波动性糖环境下对人脐静脉内皮细胞损伤的保护作用。方法:培养的人脐静脉内皮细胞用高糖和波动性糖处理48小时引起细胞损伤,采用硝酸还原酶法测定细胞培养液中一氧化氮(NO)的含量,化学比色法测定培养液中一氧化氮合酶(NOS)的活性,紫外线分光光度仪测定乳酸脱氢酶(LDH)的释放量,硫代巴比妥酸法测定丙二醛(MDA)的含量;MTT比色法检测细胞存活率;流式细胞仪分析细胞周期。结果:NAC能显著抑制高糖和波动性糖环境下人脐静脉内皮细胞NO、LDH、MDA的生成和NOS的活性及活细胞数量减少,显著减轻内皮细胞增殖的抑制。结论:NAC能减轻高糖和波动性糖环境下所致的人脐静脉内皮细胞损伤,其保护作用与抗氧化、减少自由基生成有关。 Objective: To investigate the protection of N-acetylcysteine ( NAC ) against damages of human umbilical vein endothelial cells(HUVECs) induced by intermittent glucose. Methods: Damages of cultured HUVECs were induced by treatment with high glucose and intermittent glucose for 48 hours. The release of lactate dehydrogenase (LDH) and the content of malondialdehyde(MDA) , nitric oxide(NO) and NOS activity were measured by the spectrophotometic method. The proliferation of endothelial cells was measured by MTT assay. The distribution of cells along phases of cell cycle was measured by flow cytometry. Results : NAC (4,8,16 or 32 mmol/L) markedly reduced the increased content of NO, LDH, MDA and NOS activity and attenuated the increased proliferation of endothelial ceils induced by high glucose and intermittent glucose. Conclusion: NAC can protect endothelial cells against damages induced by high glucose and intermittent glucose and the protective effect of NAC may be related to the inhibition of lipid peroxidation.
