摘要
目的探讨薯蓣皂苷元诱导人骨肉瘤U-2OS细胞凋亡的发生机制。方法用透射电镜、AO/EB染色法观察U-2OS细胞的形态学变化,用蛋白印迹法检测Caspase-8表达水平。结果经薯蓣皂苷元处理的U-2OS细胞呈典型的凋亡形态学改变,其Caspase-8蛋白表达增强。结论薯蓣皂苷元通过Caspase-8途径诱导U-2OS细胞凋亡而抑制其增殖。
Objective To explore the mechanism of human osteosarcoma U-2OS cell line apoptosis induced by diosgenin. Methods The morphological changes of U-2OS cell line induced by diosgenin were observed by transmission electron microscope and AO/EB staining, and the expression level of Caspase-8 in U-2OS cell line was detected by Western blotting. Results U-2OS cell line treated by diosgenin showed typical morphologic changes of apoptosis and the expression level of Caspase-8 increased. Conclusion Diosgenin induces human osteosarcoma U-2OS cell line apoptosis through Caspase-8 pathway to inhibit its proliferation.
