摘要
通过地塞米松体内给药建立小鼠胸腺细胞凋亡模型,普通光镜下观察凋亡细胞形态学改变并统计凋亡率,结合琼脂糖凝胶电泳检测,定性定量研究不同浓度聚乙二醇1000VE琥珀酸酯(TPGS)对胸腺细胞凋亡的影响。结果表明,TPGS能够抑制地塞米松诱导的小鼠胸腺细胞凋亡,降低凋亡率,该抑制作用呈现剂量依赖性,在低浓度(1倍需要量和5倍需要量)下最为显著,当浓度升高至10倍需要量和20倍需要量时,凋亡率与阳性对照组无显著差别。
Set up thymus cell apoptosis model by in vivo injection of dexamethasone (DEX). Observed morphological changes of cell by ordinary microscope and calculated the apoptotic rate, combined with agarose gel electrophoresis. Made a qualitative and quantitative research of the effect of D-α-to- copherol polyethylene glycol 1 000 succinate (TPGS) on thymus cell apoptosis. The results showed that TPGS inhibited thymus cell apoptosis which induced by DEX and decreased the apoptosis rate. The inhibition was dose-dependent, which means, comparing to the positive control group, 1 times and 5 times dosage were extremely significantly, when it increased to 10 times and 20 times dosage, the inhibiton became insignificant.
