摘要
目的:从胎鼠肢芽中提取对运动神经元具有生物活性的神经营养因子.方法:用Wistar大鼠第15天胎鼠肢芽制备提取液,以成年Wistar大鼠骨骼肌提取液及生理盐水为对照,在切断坐骨神经的乳鼠动物模型上,检测其运动神经营养活性.用SDS-聚丙烯酰胺凝胶电泳对两种提取液组份进行对比.结果:术后30天损伤侧相应的腰髓运动神经元存活率,注射肢芽提取液组为79.2%,成体骨骼肌提取液组为42.l%,生理盐水组为23.7%.电泳结果显示肢芽提取液比骨骼肌提取液多21KD、18KD、15KD三种蛋白组份.结论:与两对照组相比,胎鼠肢芽提取液呈现了显著的运动神经营养活性.肢芽提取液中21KD、18KD、15KD三种蛋白组份可能是其中有效的运动神经营养因子.
To prepare the extract from limb buds of embryo rats and to assay the motoneuron neurotrophic activities of the extract. Methods: The treated limb buds extract was prepared from the limb buds of 15-day-old Wistar embryo rats. The control skeletal muscles extract was pre-pared from skeletal muscles of Wistar adult rats. Physiological Saline was used for the blank con-trol. The motoneuron neurotrophic activities of the extracts were assayed in animal model of in-jured sciatic nerve of neonatal Wistar rats of 3 days. Then, the components of two extracts were analyzed by SDS-PAGE. Results: 30 dyas after resection of sciatic nerve, 79. 2% of the corre-sponding motoneurons were protected from death by multiple injection of limb bud extracts at the site of axotomy, and 42. 1% survived when treated with adult muscle extracts. Survival rate was only 23. 7% in physiological saline group. As the result of SDS-PAGE, the limb bud extract con-tained 21KD, 18KD and 15KD additional proteins as compared with the adult skeletal muscle ex-tract. Conclusions: The limb bud extract showed marked motoneuron neurotrophic activities compared with the two controls. 21KD, 18KD and 15KD additional protein components in the limb bud extract may be the effective motoneuron neurotrophic factors.
