摘要
目的:探讨人大肠癌细胞碱性成纤维细胞生长因子(bFGF)的表达、定位,并检查其受体。方法:采用免疫细胞化学染色、125I标记抗体的原位放射免疫分析及放射受体分析方法。结果:RPMI4788细胞产生的bFGF大部分蓄积于细胞核内,一部分存在于细胞膜上,并在细胞膜有其受体。结论:在肿瘤的发生发展过程中,肿瘤细胞产生的bFGF有可能起自泌性生长因子的作用,也可能直接参与核的功能。
Objective:The purpose was to investigate the expression and localization of the basic fibroblast growth factor (bFGF) and its receptors in human colon cancer cell line, RPMI 4788 .Methods:Immunocytochemistry and in situ radioimmunoassay by monoclonal antibody against bFGF labeled with 125 I were used to detect the expression of bFGF. Receptors of bFGF was detected by radioreceptorassay.Results:Most of bFGF produced by the tumor cells were accummulated in the cell nuclears and partly expressed on the cell membranes. The receptor of bFGF also existed on the same membranes.Conclusions In carcinogenesis and its progress, the bFGF produced by tumor cells may act as tumor autocrine growth factor or play important roles directly in nuclear function.
