The Effect of Nitric Oxide on Brain Edema and Regional Cerebral Blood Flow after Brain Ischemia/Reperfusion in the Wistar Rats

摘要 目的:研究NO在脑缺血再灌注中对局部脑血流量和脑水肿的影响,为临床改进脑缺血的治疗提供新思路。方法:采用拴线法大鼠MCA缺血再灌注模型,利用外源性NO前体(L-arg)和NOS抑制剂(L-NAME)。将42只Wistar大鼠随机分组,在r-CBF组和脑水肿两大组中分别设有:缺血再灌注盐水组,再灌注L-arg组,再灌注L-NAME组。结果:与盐水组比较,L-arg组r-CBF明显升高(P<0.05),脑水肿显著减轻(P<0.05);L-NAME组r-CBF明显降低(P<0.05),脑水肿显著加重(P<0.05)。结论:NO对缺血再灌注脑组织有保护性作用。 Objective: We studied the effects of Nitric Oxide (NO) on brain edema and r CBF after ischemia/reperfusion. Our purpose was to produce a new thought for improving the treatment of brain ischemia.Methods:We used Wistar rat MCA ischemia/reperfusion model, which was made by blocking the external cervical artery with thread for obstruction. The level of NO was modulated by L arginine, an exogenous precursor of NO and Nitric oxide synthase (NOS) inhibitor (L NAME).Forty two Wister rats were randomly assinged to groups.First, the r CBF group and brain edema group were determined.Then both r CBF and brain edema groups were further divided into three groups, namely:L arg, L NAME,and the control.Results:r CBF increased significantly (P<0.05) and brain edema decreased obviously (P<0.05) in L arg group as compared with those of the control. r CBF decreased sharply (P<0.05) and brain edema increased significantly (P<0.05) in L NAME group in comparison with those of the control.Conclusions:NO was protective for brain tissue in ischemia/ reperfusion.
