摘要
以提取物得率和总黄酮含量为指标,研究了超声辅助萃取条件对槐米总黄酮提取效果的影响.结果表明,超声辅助萃取条件对于槐米总黄酮提取物的得率和总黄酮含量有明显的影响,相应提取物红外光谱的特征峰型、峰高和峰位置也存在一定的差异.通过对这些提取物定量-定性分析,得出超声辅助碱溶酸沉法萃取槐米总黄酮的最佳条件为:固定超声频率20.kHz,固液比(g∶mL)1∶10,硼砂加入量为槐米质量的2%,提取液的pH为9,提取温度45.℃,超声波功率100.W,超声提取时间30.min.在此条件下,提取物的得率为18.8%,总黄酮含量为91.2%.
Using yield and flavonoids content of extracts as standards ,the ultrasound-assisted extraction process of flavonoids from the buds of Sophora japonica Linn is studied. The result indicates that the ultrasound-assisted has very strong promoting action to extraction of flavonoids from sophora rice. The FTIR spectrum of every sample is typical,with different absorption peak pattern,intensity and location,related to different extracting condition. When the ultrasonic frequency is 20 kHz and solid-to-liquid ratio is 1 : 10(g : mL) ,the optimized conditions are as follows:the mass ratio of borax to sophora rice is 1 : 50,ultrasonic power 100 W,extracting 30 min at 45 ℃ and pH 9.0. Under these conditions,the yield is 18.8% and flavonoids content is 91.2%.
