双核铜配合物对DNA的氧化切割活性

摘要 考察了两种双核铜配合物Cu2(phen)2Cl2(μ-Cl)2和[Cu2(Ophen)2].2H2O对超螺旋pBR322DNA的氧化切割活性。在两种体系条件(以抗坏血酸作还原剂而不加H2O2、不加还原剂而只加H2O2)下,双核铜配合物都表现出显著的切割活性,但前一体系的活性明显强于后者,而相同条件下配合物Cu2(phen)2Cl2(μ-Cl)2的活性强于配合物[Cu2(Ophen)2].2H2O。反应过程中产生的活性氧主要为.OH,.OH攻击糖环造成DNA链断裂,实现对DNA的氧化切割。 Oxidative DNA cleavage promoted by two binuclear copper complexes, Cu2 (phen)2 Cl2 (μ-Cl)2 and [Cu2(Ophen)2] · 2H2 O, were investigated in this paper. The two complexes exhibited efficient DNA cleavage activities at micromolar concentrations in the presence of ascorbic acid or H2O2. The system with ascorbic acid was much more efficient than that with H2O2, and under the same conditions Cu2 (phen)2Cl2 (μ-Cl)2 showed higher cleavage efficiency than [Cu2 (Ophen)2] · 2H2O. Hydroxyl radical was the main one of active oxygen species which could attack sugar ring of DNA.
