3种抗氧化剂对Cr^(6+)所致细胞生物学效应的影响 被引量:15

摘要 目的:研究维生素C、谷胱甘肽、秦美种猕猴桃汁3种抗氧化剂对铬(Cr)所致细胞毒性、DNA损伤及Cr价态的影响。方法:应用噻唑蓝比色法、单细胞凝胶电泳及原子吸收法研究Cr的细胞毒性、DNA损伤与价态的变化及3种抗氧化剂的干预作用。结果:猕猴桃汁可以有效地拮抗Cr6+引起的细胞毒性,促进Cr6+在细胞外的还原,降低Cr6+的细胞摄入,而维生素C及谷胱甘肽的作用则随Cr染毒剂量的不同而不同。结论:猕猴桃汁对Cr所致细胞生物学效应的拮抗作用明显优于维生素C及谷胱甘肽,可能与其促进Cr6+细胞外还原,阻断Cr6+细胞摄入有关。 Objective: To study the effect of Vit C and glutathione (GSH) and Qin Mei KFJ on the cytotoxicity and DNA damage and the valent state of chromium in human embryo lung(HEL) cells induced by K 2Cr 2O 7(Cr 6+ ). Methods: Thiazolyl blue tetrazolium bromide (MTT) method and single cell gel electrophoresis (SCGE) and atomic absorption were adopted to study the cytotoxicity and DNA damage and the valent state of chromium. Results: Qin Mei KFJ could antagonize the cytotoxicity induced by K 2Cr 2O 7 effectively, however, Vit C and GSH play a different role according to the different doses of K 2Cr 2O 7 used, respectively. Conclusion: Qin Mei KFJ could promote the reduction of extracellular Cr 6+ and decrease the uptake of Cr 6+ by cells, but Vit C and GSH possessed complicated action, respectively.
