摘要
目的观察17β雌二醇对原代人脐静脉内皮细胞(HUVEC)一氧化氮(NO)生成的影响。方法采用胰蛋白酶灌流消化法培养原代HUVEC,以原代HUVEC为对象,观察不同浓度的17β雌二醇(100nmol/L,30nmol/L,10nmol/L,1nmol/L,0.1nmol/L)作用于原代HUVEC一定时间下NO产量(测代谢产物NO-2),采用Greis反应测定NO-2的量。结果100nmol/L17β雌二醇作用1小时,NO-2即明显增高(P<0.001);30nmol/L的17β雌二醇作用下,4小时起NO-2产量即明显增高(P<0.001);10nmol/L的17β雌二醇作用12小时及1nmol/L的17β雌二醇作用18小时,NO-2产量均有明显增高(P<0.05)。0.1nmol/L的17β雌二醇作用24小时内,NO-2产量与对照组比较无统计学差异(P>0.05)。17β雌二醇作用的阈浓度可能在0.1nmol/L-1nmol/L之间。结论17β雌二醇可促进原代人脐静脉内皮细胞NO的生成,可能为其抑制动脉粥样硬化形成、降低血管阻力的重要机理之一。
Objective To investigate the effects of estrogen on the production of nitric oxide (NO). Methods The primary cultures of human umbilical vein endothelial cells (HUVEC) were harvested by trypsin. Confluent primary cultures of HUVEC were exposed to 17 β estradiol in medium. The production of NO was assessed by Griess reaction. Results Exposure of the cells to 17 β estradiol (1nmol/L, 10nmol/L, 30nmol/L, 100nmol/L) enhanced the production of NO at 18 24 hours, 12 24 hours, 4 24 hours, 1 24 hours respectively ( P <0.01), however, the effect was not observed with 17 β estradiol at 0.1nmol/L within 24 hours.Conclusion 17 β estradiol could enhance the production of NO from the primary cultures of HUVEC. This finding suggests the antiatherosclerotic effect and the reduction of vessel resistance of estrogen.
