儿童外周血成纤维细胞集落培养方法初探

A Preliminary Exploration for the Method of CFU-F Culture in the Peripheral Blood of Children

取健康儿童无菌外周血，用1.119g／ml淋巴细胞分离液分离细胞，制成单细胞悬液进行培养，培养开始时加入1，25双羟基维生素D3和碱性成纤维细胞生长因子，刺激促进成纤维前体细胞分化增殖，形成成纤维细胞集落，建立了培养方法。为儿童血循环中存在间充质干细胞的假说提供了初步实验依据，并为儿童骨骼、肌肉营养代谢性疾病和造血系统疾病研究提供了新的途径。

Human peripheral blood cells were obtained from two healthy childtgh donors. mhoheral blood monouclear cells and granulocytes were separated by centrifugation over a ficoll - hypaque density gradient(1. 119g/ml), suspended in the specific culture medium supplemented with 30% FCS, then plated in the flasks. With the addition of 1, 25 - dihydroxyvitaminD3 and basic fibroblast growth factor to enhance the differentiation of fibroblast - precursor and the proliferation of fibroblast, colony forming units - fibroblast (CFU - Fs) were formed. This result is the preliminary evidence for the hypothesis that the fibroblast - precursors are circulating in the peripheral blood of children. and this culture methed provides a new approach to the study of the nutritional and metabolic diseases of skeleton and muscles and haemopoietic diseases.
