Expression and significance of Tiam1 in breast cancer patients

摘要 目的探讨Tiam1在正常乳腺组织、乳腺癌组织及不同侵袭能力乳腺癌细胞系中的表达情况。方法应用免疫组化方法检测Tiam1在60例乳腺癌组织和34例正常乳腺组织中的表达。应用Westernblot方法检测Tiam1在乳腺癌细胞系MDA-MA-231(高侵袭)和MCF-7(低侵袭)中的表达。结果Tiam1阳性率在正常乳腺组织(41%)中明显低于乳腺癌组织(71.7%,P<0.01),Tiam1在乳腺癌组织中的表达与肿瘤的TMN分期、是否发生淋巴结转移以及病理分级密切相关(P<0.05)而与肿瘤的大小无关。Tiam1总蛋白在乳腺癌高侵袭细胞系中的表达量高于低侵袭细胞系,差异具有显著性(P<0.05)。结论Tiam1在乳腺癌中的表达与其转移侵袭相关,可能是判断预后的一个潜在指标。 Objective To investigate the expression of Tiaml in normal breast,breast carcinoma tissue,and cell lines with different invasive abilities. Methods The expression of Tiaml in 60 specimens of breast carcinoma and 34 specimens of normal breast tissues was detected by SP immunohistochemistry. The expression of Tiaml in breast cancer cell lines, MDA-MB-231 with high invasive ability and MCF-7 with low invasive ability,was detected by Wes tern blot. Results Tiaml was expressed in cell cytoplasm in normal breast tissues and breast carcinoma. The cytoplasmic positive rates of Tiaml were 41% in normal breast tissues,71.7% in breast carcinoma. The positive rate of Tiaml was significantly lower in normal breast tissues than in breast carcinoma (P〈0.01).The levels of protein of Tiaml were significantly related with the lymph node metastasis, different clinical stage and histological grade(P〈0.05 ), but no correlation with tumor size (P〉0.05). The protein level of Tiaml was higher in MDA-MB-231 cells than in MCF-7 cells (P〈0.05). Conclusion Tiaml gene plays an important role in invasion and metastasis of the breast carcinoma, these results strongly suggested that the expression level of Tiaml couold indicate the potential indicator for diagnosis of metastasis to some extent.
