摘要
目的探讨养肺活血方对博莱霉素诱导肺纤维化大鼠肺组织细胞外信号调节激酶1/2 (ERK1/2)和核因子κ -B(NFκ-B)表达的影响。方法大鼠麻醉后,经气管软骨环间隙向心端穿刺,气管内注入博莱霉素A5 (5 mg/kg)建立大鼠肺纤维化模型,假手术组大鼠在同样麻醉条件下气管内注入等量生理盐水。术后第2日开始每日灌胃给药,假手术组和模型组予5‰羧甲基纤维素钠(CMC钠)溶液,地塞米松组予等容积地塞米松蒸馏水溶液,各中药治疗组予等容积相应中药合剂,灌胃28 d,麻醉处死大鼠,将左肺固定,包埋切片,常规脱蜡。用免疫组化SABC法检测ERK1/2和NFκ-B的表达;采用光学显微镜和全自动图像获取系统随机测量并计算平均光密度(I OD)值。结果模型组大鼠肺组织ERK1/2和NFκ-B的表达明显高于假手术组(P<0.01)。各中药治疗组肺组织ERK1/2和NF κ-B的表达明显低于模型组(P<0.01)。结论影响ERK1/2和NFκ -B信号通路可能是中药养肺活血方治疗肺纤维化的机制之一。
Objective To study the effect of Yangfei Huoxue Prescription on ERK1/2 and NFκ-B of bleomycin-induced pulmonary fibrosis in rats. Methods All of rats were randomly divided into seven groups and were intratracheally infused with bleomycin to build the model. Sham operation group and model group were given 5%0 CMC, dexamethasone group was given dexamethasone solution, and each treatment group was given corresponding Chinese medicine mixture. After histological process, the expression of ERK1/2 and NFκ-B were examined by SABC immunohistochemical method. Results Expression of ERK 1/2 and NFκ-B in model group were higher than that in sham operation group (P〈0.01). Yangfei Huoxue Prescription could obviously decrease the degree of expression of ERK1/2 and NFκ-B of Bleomycininduced pulmonary fibrosis in rats (P〈0.01). Conclusions Yangfei Huoxue Prescription can decrease the degree of ERK1/2 and NFκ-B in lung, which may be one of the mechanisms in treating pulmonary fibrosis.
